One Of The First Studies To Look At Human Cells
The researchers used stem cells from patients with Parkinsons disease who had a mutation in the gene responsible for encoding the -synuclein protein.
At least 30 alterations in this gene have been associated with Parkinsons, and -synuclein protein clumps are a well-documented, albeit poorly understood, hallmark of the disease.
For the new research, the scientists also worked with normal embryonic cells that they modified using genetic editing to replicate the -synuclein genetic mutation.
Prof. Ryan explains why using human cells makes this study particularly valuable. Until now, he says, the link between pesticides and Parkinsons disease was based primarily on animal studies as well as epidemiological research that demonstrated an increased risk among farmers and others exposed to agricultural chemicals.
We are one of the first to investigate what is happening inside human cells, explains Prof. Ryan.
Stem cells are undifferentiated cells that go on to individualize into specific types of cells. Prof. Ryan and his colleagues used the two types of stem cells to derive dopamine-producing nerve cells from them.
Then, they exposed these dopaminergic neurons which are known to be affected the most by Parkinsons disease to the two pesticides.

Nih Study Finds Two Pesticides Associated With Parkinsons Disease
New research shows a link between use of two pesticides, rotenone and paraquat, and Parkinson’s disease. People who used either pesticide developed Parkinsons disease approximately 2.5 times more often than non-users.
The study was a collaborative effort conducted by researchers at the National Institute of Environmental Health Sciences , which is part of the National Institutes of Health, and the Parkinson’s Institute and Clinical Center in Sunnyvale, Calif.
“Rotenone directly inhibits the function of the mitochondria, the structure responsible for making energy in the cell,” said Freya Kamel, Ph.D., a researcher in the intramural program at NIEHS and co-author of the paper appearing online in the journal Environmental Health Perspectives. “Paraquat increases production of certain oxygen derivatives that may harm cellular structures. People who used these pesticides or others with a similar mechanism of action were more likely to develop Parkinson’s disease.
There are no home garden or residential uses for either paraquat or rotenone currently registered. Paraquat use has long been restricted to certified applicators, largely due to concerns based on studies of animal models of Parkinson’s disease. Use of rotenone as a pesticide to kill invasive fish species is currently the only allowable use of this pesticide.

ALDH increases the dopamine metabolite 3,4-dihydroxyphenylacetaldehyde, which produces degeneration among neurons associated with the production of dopamine. One of the central dopamine-producing centers exists in the brain the substantia nigra located within the midbrain.
When the nerve cells located in this region die off or become otherwise deranged, they stop producing dopamine and other neurotransmitters that help control coordination and movement throughout the body. A lack of these neurotransmitters will produce the shakiness and eventual loss of coordination characteristic amongst progressed Parkinsons patients.

Occupational exposure to the herbicide paraquat was not found to be associated with a greater risk of Parkinson disease or increased risk of mortality related to other causes.
Amid ongoing legal proceedings examining paraquat exposure with risk of Parkinson disease , a recent study published in the Journal of Occupational Medicine and Toxicology indicates that the herbicide has no association with increased risk of PD or death from any cause.
With research over the past 35 years having evaluated environmental factors, including rural living, well-water consumption, farming, and pesticide exposure with risk of PD, researchers highlight that notable interest has focused on the quaternary ammonium herbicide PQ due to its structural similarity with a toxin, called 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine , that has been shown to cause permanent parkinsonism in humans.
Many experimental animal and in vitro studies of PQ have been performed looking for effects, such as deficits in dopaminergic neurons, but there remain many challenges in interpreting the relevance of these studies to humans, said the study authors.
Conducting an extended follow-up of 7.5 years, health data of 926 male and 42 female workers derived from the UK National Health Service digital database were included in the retrospective cohort analysis. Mortality rates for males were compared with national and local rates, including rates for PD as a mentioned cause of death.

The Environmental Protection Agency has found evidence linking exposure to paraquat dichloride, an herbicide colloquially known as paraquat, to the development of Parkinsons disease later in life, per a 2016 regulatory filing. Now, agricultural workers who have been exposed to this toxic chemical may have the right to file lawsuits against paraquat manufacturers for financial compensation.
As reported by the National Law Review, many countries have effectively banned paraquat due to its high level of toxicity. What Is The Causal Effect Between Paraquat Herbicide And Parkinsons
Paraquat may have been considered the holy grail of herbicides cheap and potent in the 60s and even later, but in time, more and more studies have demonstrated the undeniable and tragic link between paraquat and Parkinsons disease. Unfortunately, unwanted grass and weeds were not the only things that paraquat destroyed. Paraquat has a similar chemical structure with an active metabolite called 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine . This metabolite is shown to cause lesions in the substantia nigra, pars compacta region of the brain in rats, leading to Parkinsons disease.
According to the Yonsei Medical Journal, it seems that paraquat accounts for 20 deaths per million people. Rotenone Linked To Parkinsons
A study from Koreas Yonsei University studied the broad spectrum pesticide Rotenone and how it damages nerve cells and pathways.
The researchers found that Rotenone induces cell death in a process called with G2/M cell cycle arrest. G2/M cell cycle arrest blocks the process of mitosis that enables cells and their DNA to replicate and more importantly among nerve cells repair any DNA damage.
Thus the insecticide basically blocks the ability of the nerve cell to repair itself lending to the cells eventually dying off or mutating.

Rural Living And An Increased Risk Of Pd
In the 1980s, studies were conducted that showed that early-age exposure to a rural environment as well as exposure to well water were associated with development of PD later in life. Subsequently, multiple additional studies looked at these questions. The studies are mixed in their conclusions, but overall the evidence supports associations between increased PD risk with each of the following:
- farming as an occupation,
- well water drinking, and
- living in a rural area.
Of course, all these categories are inter-related, since farmers live on farms in rural areas, are exposed to farm animals, are more likely than urban dwellers to drink well water and use pesticides. The studies were attempting to tease out why rural environments increased the risk of PD. Do only those who actually farm have an increased risk or is it enough to live on a farm? Is pesticide exposure the reason for the increased risk? Well water exposure? Exposure to farm animals? Or is it another element of rural life?
In the end, epidemiologic data supports the assertion that each of these elements increases the risk of PD. Of note, all of the increased risks in these studies are small on the order of 1.5-2 times the risk of the general population.

Exposure To Pesticides In The Military
Agent Orange was an herbicide that US troops sprayed in Vietnam from 1961-1971 to kill trees and crops that provided protection and food to the rival army. It is a mixture of two chemicals: 2,4-Dichlorophenoxyacetic acid and 2,4,5-Trichlorophenoxyacetic acid. Agent Orange was also contaminated with Dioxin, a chemical even more damaging than Agent Orange itself, since it is very long-lasting.
The effects of Agent Orange on both the Vietnamese population and on American soldiers has been studied extensively, but with much variability in the results. Birth defects have been attributed to Agent Orange exposure, as well as multiple types of cancer.
With the understanding that the Veteran community served selflessly on behalf of the American people and therefore deserve the protection and support of the American government, the Agent Orange Act was passed in 1991, allowing the Department of Veteran Affairs to declare certain conditions presumptive to exposure to Agent Orange, even if the scientific data associating Agent Orange with that condition was not airtight.

No One Definitive Cause Of Parkinsons
There are no biomarkers or objective screening tests that indicate one has Parkinsons. That said, medical experts have shown that a constellation of factors are linked to it.
Parkinsons causes are likely a blend of genetics and environmental or other unknown factors. About 10 to 20 percent of Parkinsons disease cases are linked to a genetic cause, says Ted Dawson, M.D., Ph.D., director of the Institute for Cell Engineering at Johns Hopkins. The types are either autosomal dominant or autosomal recessive .
But that leaves the majority of Parkinsons cases as idiopathic, which means unknown. We think its probably a combination of environmental exposure to toxins or pesticides and your genetic makeup, says Dawson.
Age. The biggest risk factor for developing Parkinsons is advancing age. The average age of onset is 60.
Gender. Men are more likely to develop Parkinsons disease than women.
Genetics. Individuals with a parent or sibling who is affected have approximately two times the chance of developing Parkinsons. Theres been an enormous amount of new information about genetics and new genes identified over the past 10 or 15 years that have opened up a greater understanding of the disease, says Dawson.
