Caffeine May Modulate Pd Pathology By Regulating Autophagy Activity

Using an -Syn fibril model of PD, we recently have provided the first evidence that caffeine can attenuate abnormal -Syn aggregation and neurotoxicity by re-establishing autophagy activity in animal models of PD . Specifically, chronic caffeine treatment did not affect autophagy processes in the normal mice striatum, but did selectively reverse -Syn-induced defects in macroautophagy and CMA . Thus, caffeine may represent a novel pharmacological therapy for PD by targeting autophagy pathway. This study collaborates with the previous study showing that caffeine-induced autophagy protected against human prion protein peptide -triggered apoptosis in a SH-SY5Y neuroblastoma cell line. Therefore, autophagy enhanced by caffeine may be a valid therapeutic strategy for neurodegenerative diseases such as PD and prion .

Mounting evidence is indicating the role of mitochondrial dysfunction in the pathogenesis of PD . Though motor deficit is the major outcome of PD, several pathological events, including mitochondrial dysfunction, occur earlier before the clinical symptoms become prominently visible. Normally, -synuclein gets localized in mitochondrial associated membranes, thereby forming an interface between mitochondria and endoplasmic reticulum that helps in the regulation of Ca2+ signaling and apoptosis. An increase in the expression of -synuclein or the presence of its aggregated form causes dissociation of mitochondria and ER, which further results in the loss of Ca2+ signaling and decrease in mitochondrial energy production . Case Ascertainment And Follow

We identified potential Parkinson’s disease cases from three independent sources.

  • Follow-up interviews were conducted with surviving cohort subjects between 1999 and 2004, on average 7 years after enrollment. The response rate was 90 percent. Subjects were asked whether they had ever been told by a physician that they had Parkinson’s disease and, if yes, age at which diagnosis was ascertained.

  • Computer linkage of the cohort database with the nationwide hospital discharge database was carried out. This database was set up in 1990 by the Singapore government to capture all inpatient discharge information nationwide . We identified all diagnoses accompanied by International Classification of Diseases, Ninth Revision code 332 and code 3320 .

  • Computer linkage of the cohort database with two hospital-specific Parkinson’s disease registries in Singapore was carried out.
